Abstract

Background: Cholesterol is a fatty substance circulating in the blood, produced by the liver. To check cholesterol levels, a sample in the form of serum is needed. The reality in the field during clinical chemistry examinations is that there are differences in treating blood samples, after taking venous blood samples, they are immediately centrifuged without being frozen in order to shorten the time. Objective: To know the total cholesterol levels after taking 15 minutes, 30 minutes and 45 minutes before being centrifuged. Methods: This type of research is experimental with 40 research samples of UPT employees. Sriwijaya Indralaya University Health Clinic, This examination uses the CHOD-PAP method. Venous blood collected in the vacutainer tube after taking 15 minutes, 30 minutes and 45 minutes was then centrifuged and processed into serum, then the total cholesterol results were checked with a chemistry analyzer (Humstar 100). The results of the total cholesterol examination were analyzed by the Anova test with a 95% confidence level. Results: The average total cholesterol level after taking 15 minutes before being centrifuged was 165.73 mg/dL, after taking 30 minutes before being centrifuged 155.78 mg/dL. dL, while after 45 minutes before being centrifuged it was 164.45 mg/dL. Conclusion: There are differences in the results of examination of total cholesterol levels after taking 15 minutes, 30 minutes and 45 minutes before being centrifuged, obtained with a P-value of 0.013 <0.05, there is a significant difference. Blood serum total cholesterol levels after 15 minutes of collection tend to be higher than blood serum cholesterol levels after 30 minutes and 45 minutes before being centrifuged Suggestion: To check total cholesterol, blood should be allowed to freeze before centrifuging to obtain perfect serum perform an examination with other fat component variables.
