Abstract

Background: Examination for the liver and heart is an emergency, one of which is ALT examination, so a rapid examination must be carried out. To speed up the inspection can be done by reducing the centrifugation time. Reducing the centrifugation time to 1 minute at a speed of 5000 rpm is expected to give the same results as centrifugation for 5 minutes at a speed of 3000 rpm, so that the ALT examination time can be faster, shorten patient waiting time, and save electricity by reducing variable costs. Objective: This study aims to determine whether there is a difference in serum ALT levels which were centrifuged at 3000 rpm for 5 minutes and 5000 rpm for 1 minute. Methods: This research is a pre-experimental research with Static Group Comparison research design. Examination using the IFCC method without pyridoxal. The research subjects were 30 people. The sample was serum from blood which was frozen for 30 minutes and then centrifuged at 3000 rpm for 5 minutes and 5000 rpm for 1 minute. Samples were examined using the Biosystem A15 tool. Data were analyzed using Paired T Test. Result: The results showed that the average value of ALT levels centrifuged at 3000 rpm for 5 minutes was 19.27 U/L and at 5000 rpm for 1 minute was 18.70 U/L. Based on the Paired T Test, there was no difference in ALT levels in serum from blood that was centrifuged at 3000 rpm for 5 minutes and 5000 rpm for 1 minute, p-value (sig) = 0.188 with a significant level (α = 0.05), then p -value (sig) > 0.05. Conclusion: There was no difference in serum ALT levels from blood centrifuged at 3000 rpm for 5 minutes and 5000 rpm for 1 minute. Suggestion: Centrifugation at a speed of 5000 rpm for 1 minute can be considered as an alternative procedure in processing specimens for ALT examination in the laboratory.
