Abstract

Background: Examination of erythrocytes is carried out as a routine screening part of a complete blood count and routine blood tests to diagnose hematological disorders. The important thing in the pre-analytic stage is homogenization. Blood in a tube containing an anticoagulant must be mixed immediately to prevent clotting. The delay in the examination causes the blood to precipitate, so secondary homogenization is necessary before the blood is examined so that the blood is normally distributed. Primary homogenization was carried out 10 times, but there is no standard for secondary homogenization yet. Objective: Knowing the results of the erythrocyte count using the secondary homogenization technique 5 and 8 times after being left for 60 minutes. Method: This type of research is pre-experimental with Static Group Comparison and with total sampling technique.All samples were primary homogenized 10 times, then allowed to stand for 60 minutes, after which the samples were secondary homogenized 5 times and 8 times.Erythrocyte levels were checked using Sysmex KX-21N. Data were analyzed using the Wilcoxon test with a 95% confidence level. Result: Examination of secondary homogenized erythrocytes 5 times obtained a median value of 461 X106µ, a minimum value of 42 X106µL and a maximum value of 586 X106µL. While the secondary homogenized 8 times, the median value was 456 X106µL, the minimum value was 47 X106µL and the maximum value was 588 X106µL. Statistical test results showed no significant difference p = 0.416 (> 0.05). Conclusion: Based on the results of the study, it was concluded that there were no differences in erythrocytes using the secondary homogenization technique 5 times and 8 times. Suggestion: Processing of specimens in performing secondary homogenization can be done with 5 times homogenization.
