Abstract

Natalia, NIM : 1534010, The different numbers of Trichophyton rubrumcolonies on Sabouraud Dextrose Agar (SDA) culture medium and its modification using 3 gram of glucose.bachelor Thesis of DIV Medical Analysis, Faculty of Medical Science, Universitas Katolik Musi Charitas Palembang. Background :The laboratory analysis to diagnose the etiology of Trichophyton rubrumcan be done using macroscopy, microscopy, serology, biomolecular analysis, biopsy, and culture examination. Culture examination is the gold standard to analyze or examine the fungus microorganism. In culture examination, Sabouraud Dextrose Agar (SDA) can be used as the culture medium. However, the low fertility rate becomes the common problems to regrow the culture on SDA medium. In this study, the low fertility rate on SDA medium can be helped by providing an additional 3 grams of glucose to fulfill the nutrition in the process of growing fungal colonies in fungal culture examination. Method : This research was categorized as a true experimentPosttest-only control design. All the experiment was conducted in Microbiology Laboratory of Faculty of Medical Science, Universitas Katolik Musi Charitas Palembang. The main subject investigated in this research was Trichophyton rubrumwhich was growth on SDA medium and SDA medium with an addition of 3 grams Glucose. The numbers of inoculum were calculated by measuring the inoculum optical turbidity of Trichophyton rubrumusing a McFarland standard of 0,5. The inoculum was prepared by diluting the medium to 102. The numbers ofTrichophyton rubrumwas then calculated on the sixth day after the inoculation, and all the obtained data were further analyzed using Paired T-test. Result : The result showed that the colonies number of Trichophyton rubrumafter day sixth was 142 CFU/mL on Sabouraud Dextrose Agar (SDA) medium without adding any glucose. Furthermore, providing additional glucose (3 grams) enhances the colonies numbers in which 204 CFU/mLTrichophyton rubrumobtained after six incubation days. Pairt-test was then confirmed that providing additional glucose on SDA medium provided a significant enhancement by showing p values of 0.001 with a significant levelof 0.05 (p<α). Conclusion : There was a significant effect of glucose on Sabouraud Dextrose Agar (SDA)medium in enhancing the colonies number of Trichophyton rubrum.
