Abstract

Background: Antibiotic susceptibility testing is important for determining the effectiveness of antibiotics against bacteria that cause infections, including E. coli. The Kirby-Bauer disk diffusion method uses standard antibiotic disks as a reference, but the use of injectable ceftriaxone impregnated into filter paper has the potential to be a more economical alternative. Previous studies have shown that the inhibition zone diameter of injectable ceftriaxone is still smaller than that of standard discs; therefore, a study is needed to examine the effect of increasing the concentration of injectable ceftriaxone on the inhibition zone diameter of E. coli. Objective: To determine the effect of variations in the concentration of injectable ceftriaxone on the inhibition zone diameter of E. coli Methods: A pre-experimental study using a Static Group Comparison design. The experiment was conducted at the Microbiology Laboratory of the Faculty of Health Sciences Musi Charitas Catholic University Palembang. The samples consisted of pure E. coli ATCC 25922 strains treated with ceftriaxone. Based on Federer’s formula, each treatment was repeated 10 times. Data were analyzed using the One-Way Anova test followed by the Tukey post hoc test. Results: The average diameter of the inhibition zone for the ceftriaxone disk was 32.93 mm; for ceftriaxone injection at concentrations of 35 µg, 20.4 mm; 40 µg, 24.07 mm; and 45 µg, 26.48 mm. A one-way ANOVA test yielded p = 0.000. Tukey’s post hoc test for 35 µg: p = 0.000; 40 µg: p = 0.000; and 45 µg: p = 0.000 Conclusion: There was an effect of concentration variation on the diameter of the inhibition zone of E. coli using ceftriaxone discs and ceftriaxone injections (p=0.000). Suggestion: Further research is recommended to investigate higher concentrations in order to align the results with those of standard disks and to use blank paper disks as a substitute for filter paper.
