Abstract

The Erythrocyte Sedimentation Rate (ESR) is a test used to measure the rate at which erythrocytes settle in non-clotted blood (blood containing an anticoagulant) within a vertical tube over a specific period. One factor influencing the ESR value is the balance between blood volume and anticoagulant volume. Excessive blood volume leads to erythrocyte morphological changes such as crenation (shriveling of erythrocytes), whereas insufficient blood volume causes hemodilution (blood dilution resulting from increased plasma volume). To determine ESR values from blood samples using variation volumes 1.5 mL (1:3 ) and 3 mL (1:1 ) in K2EDTA tubes via an automated method. This study employed an observational analytic design with a cross-sectional approach. The subjects were 35 female students from the Medical Laboratory Technology (Diploma IV) program at the Faculty of Health Sciences, Musi Charitas Catholic University Palembang. The research was conducted at The public Health Laboratory Center Palembang. Statistical analysis was performed using the Wilcoxon signed-rank test. The mean ESR result for the 1.5 mL K2EDTA blood volume was 18.37 mm/hour, while the mean ESR result for the 3 mL K2EDTA blood volume was 18.51 mm/hour. There was no significant difference in ESR results between the 1.5 mL and 3 mL K2EDTA blood volumes (p-value = 0.724; >0.05).
