Abstract

Background: Blood group testing using the tube method is the gold standard for ensuring the accuracy of blood transfusions. Variations in antiserum volume are thought to influence the degree of agglutination; however, there is currently no evidence regarding the most efficient antiserum volume capable of producing an optimal degree of agglutination in a 5% erythrocyte suspension. Objective: To determine differences in the degree of agglutination during blood group testing on 5% blood group suspension samples using the tube method Methods: This study was an analytical observational study with a cross-sectional design. The study subjects were 34 students from the Diploma IV Programme in Medical Laboratory Technology. Data were analysed using the Kruskal-Wallis test and Dunn’s post-hoc test Results: From the 34 samples with three different treatments, the results showed that testing using antiserum volumes of 1, 2 and 3 drops in a 5% erythrocyte suspension sample using the tube method yielded an average agglutination degree of 4 positive (4+) Conclusion: There was no difference in the degree of agglutination in blood group testing using a 5% erythrocyte suspension with the tube method and antiserum volumes of 1, 2 and 3 drops, with a p-value of 1.000 > 0.05. Recommendation: The use of a 1-drop (50 µl) volume of antiserum is sufficient to produce a clear degree of agglutination in tube-based blood group testing using a 5% erythrocyte suspension, thereby potentially improving reagent efficiency, saving time and producing accurate results.
