Abstract

Background: Human Chorionic Gonadotropin (HCG) is a hormone synthesized by the placenta from the early stages of pregnancy and can be detected in urine. One method for detecting HCG is the immunochromatographic strip test, which can be used semi-quantitatively through serial dilution. Delayed examination is a common occurrence in laboratory practice and may cause changes in urine components. These changes are suspected to affect HCG stability and potentially influence the results of semi-quantitative urinary HCG measurement. Objective: To determine the differences in semi-quantitative urinary HCG levels at different examination delay times. Methods: This pre-experimental study employed a non-probability sampling technique with purposive sampling. The study samples consisted of urine from second-trimester pregnant women at Klinik Bidan Griya Bunda Ceria Palembang, with a total of 32 samples. Each sample was examined immediately after collection, after a 1-hour delay, and after a 2-hour delay. Data were analyzed using SPSS version 22 with the Kruskal–Wallis test. Results: The mean urinary HCG levels were 23,750 mIU/mL for immediate examination, 23,000 mIU/mL after a 1-hour delay, and 20,875 mIU/mL after a 2 hour delay. The Kruskal–Wallis test showed a significance value of 0.416 (p > 0.05). Conclusion: There was no significant difference in semi-quantitative urinary HCG levels between immediate examination and examinations delayed by 1 hour and 2 hours. Recommendation: A delay of up to 2 hours may still be considered acceptable. Further studies are recommended using quantitative methods, evaluating additional factors affecting HCG stability during delayed examination, and investigating longer delay intervals.
