Abstract

The tube method is considered the gold standard in blood grouping tests because it produces accurate and clearly observable agglutination reactions. One crucial factor influencing the success of this method is the quality of the red blood cell (RBC) suspension. In practice, a 10% RBC suspension is not always used immediately after preparation but is often stored beforehand. However, the effect of 24-hour storage on agglutination results has not been extensively studied. This study aimed to determine the differences in agglutination grades in blood grouping using the tube method with 10% RBC suspension examined immediately and after 24 hours of storage at 2–8°C. The research was an analytical observational study with a cross-sectional approach. Samples consisted of 10% RBC suspensions prepared from 31 undergraduate Medical Laboratory Technology students. Examinations were carried out immediately and after 24 hours of storage. Data were analyzed using the Wilcoxon Signed Rank Test. The agglutination grade for both immediate and 24-hour examinations was consistently 4+ across all samples. The Wilcoxon test showed a p-value of 1.000 (p > 0.05), indicating no significant difference between the two time points. The 10% RBC suspension remained stable and produced optimal agglutination results even after 24 hours of storage at 2–8°C. Short-term storage did not significantly affect antigen reactivity. Thus, 10% RBC suspensions stored for up to 24 hours can be reliably used in tube method blood grouping as an efficient practice without compromising accuracy.
