Abstract

Background : Staphylococcus aureus is a Gram(+) coccus, -hemolytic, sensitive to novobiocin, and is one of the 10 leading infectious causes of death in the world. Erythromycin is one of the recommended antibiotics in antimicrobial susceptibility testing for Staphylococcus aureus. Staphylococcus aureus grows well on medium containing blood or serum. Staphylococcus aureus antimicrobial susceptibility test is recommended using Mueller Hinton Agar medium because it gives the best results for Staphylococcus aureus Method : This research is true experimental. The research sample was a pure strain of Staphylococcus aureus bacteria which was suspended and the turbidity was matched to the MacFarland standard 0.5. The test was carried out on one medium with two treatments, namely Mueller Hinton Agar which was stored at 2-8̊C and Mueller Hinton which was stored at 20-25̊C, with 20 plates each. Inoculation of the medium using the Spread plate method was then incubated at 37ºC for 24 hours. Data were analyzed using Paired T-Test test Results : The average inhibition zone produced by Staphylococcus aureus on Mueller Hinton agar medium stored at 2-8̊C was 26.5 mm and the average for Mueller Hinton medium stored at 20-25̊ was 26.5 mm. The results of the paired T-Test showed that there was no difference in the zone of inhibition of Staphylococcus aureus against Erythromycin on Mueller Hinton Agar medium stored at 2-8̊C with Mueller hinton agar stored at 20-25̊C by looking at the probability (Sig.) of 0.083 >0.05. Conclusion : Based on the research, it can be concluded that there is no difference in the inhibition zone of Staphylococcus aureus bacteria to erythromycin on Mueller Hinton Agar medium stored at 2-8̊C with Mueller hinton agar stored at 20-25̊C.
