Abstract
Background : Cholesterol is an essential parameter in clinical chemistry testing. Its stability can be affected by pre-analytical processes, including the timing of serum separation after centrifugation. Delays in separation may allow ongoing cellular metabolism, potentially altering serum composition, including cholesterol levels. Objective : This study aimed to determine the difference in cholesterol levels between serum separated immediately and serum separated 85 minutes after centrifugation.. Methods : This type of research is pre-experimental with static group comparison design. The research sample was 38 DIV Medical Laboratory Technology students who met the inclusion and exclusion criteria. Cholesterol level examination was carried out using the CHOD-PAP method with the Biolabo Kenza 450 TX tool. Data were analyzed statistically using the Wilcoxon test. Results : The average cholesterol level in the serum separated immediately was 169,612 mg/dL, while in the serum separated 85 minutes after centrifugation it was 172,626 mg/dL. The Wilcoxon test results showed a p value = 0.001 which means there is a significant difference between the two groups. Conclusion : There is a significant difference in cholesterol levels between immediate and 85-minute delayed serum separation. Delayed serum separation may lead to a decrease in cholesterol levels.
