Abstract
Background: To determine the inhibition of Fluconazole against Candida albicans, it is necessary to test the inhibition of the fluconazole. The antifungal inhibition test against this fungus was based on measuring the anti fungal ability to inhibit the growth of fungi in vitro. The culture media used for the growth of Candida albicans were Saboroud Dextrose Agar and Mueller Hinton Agar.Saboroud Dextrose Agar medium is selective medium for fungal growth, while Mueller Hinton Agar medium is general medium for testing the sensitivity of antimicrobial compounds. This study aims to determine the difference in the inhibition zone of fluconazole using Mueller Hinton Agar and Saboroud Dextrose Agar test media. Objective: It is known that difference in the fluconazole inhibition zone on the Mueller Hinton Agar and Saboroud Dextrose Agar test media is known. Methods: This research is a quantitative research with an analytical abservational research design that is a cross sectional approach. The sample in this study was the Candida albicans strain ATCC 90028. Candida albisans colonies were inoculated in NaCl solution with a turbidity of 0,5 McF, then spread on the test medium Mueller Hinton Agar and Saboroud Agar and fluconazole disc was offixed to the top of the sheet. After incubation, the diameter of the inhibition zone formed was measured. The results of the measurement of the diameter of the inhibition zone were analyzed by paired T-test with a 95% confidence level. Results: The average diameter of the inhibition zone produced by fluconazole against Candida albicans on Mueller Hinton Agar medium was 30,5 mm and the average inhibition zone of fluconazole aganst Candida albicans ao Saboroud Dextrose Agar medium was 30,5 mm. Conclusion: There was no difference in the results of the diameter of the inhibition zone using Mueller Hinton Agar and Saboroud Dextrose Agar medium with a value of (sig 2 tailed) 0,666>0,05. Suggestion: Mueller Hinton Agar medium can be used as a substitute for Saboroud Agar Medium in a fluconazole inhibition test against Candida albicans. Further research can be carried out to see the comparison of inhibition zones by using fungal growth medium and other antifungal susceptibility tests.
