Abstract
Background : : One of the liver function tests in the laboratory is the ALP test using blood specimens that are processed into a serum. Blood specimens are taken using a syringe. The use of syringes in an inappropriate way can increase the risk of in vitro hemolysis which is mild hemolysis due to errors in the pre-analytic phase when taking the specimen, and the specimen is still used. A technical error in the pre-analytic phase is when the blood is removed from the syringe and put into a tube with strong pressure without releasing the needle, which causes red blood cells (erythrocytes) to rupture. Objective : The difference in ALP levels between hemolyzed serum 1 g/L and non-hemolyzed serum. Method : This research is pre-experimental research with One Group Pretest-Posttest research design. The test applies IFCC kinetic method. The research subjects were 36 people. The samples were serum from blood, which were left for 30 minutes and then centrifuged at 3000 rpm for 5 minutes. The samples were examined using Kenza 450 TX. The data were analyzed using paired t test. Results : The results show that the average ALP level in non-hemolyzed serum is 101.4 U/L and 106.8 U/L in hemolyzed serum 1 g/L. The results statistically show a significant difference (p=0.028) in ALP levels between non-hemolyzed and hemolyzed serum 1 g/L by applying the IFCC kinetic method. Conclusion : Based on the results of the research, it can be concluded that there is a significant difference in the test results of ALP levels in non-hemolyzed and hemolyzed serum 1 g/L by applying the IFCC kinetic method with a value of (Sig 2 tailed) 0.028 < 0.05. Suggestion : The sample in used the ALP test by ATLM to enforce diagnosis is non- hemolyzed serum. For further researchers, further research can be carried out regarding the examination of AST by using hemolyzed serum below of 1 g/L, which is 0.50 g/L and compared with non-haemolyzed serum and before measuring serum pH first.
