Abstract

Background : Aspartate aminotransferase (AST) is a mitochondrial enzyme that functions to catalyze of the reversible transfer of amino group from aspartic acid to alpha oxaloacetic acid to glutamic acid and oxaloacetic acid. The use of a syringe in withdrawal the blood can increase the risk of in vitro hemolysis, hemolysis can occur when the blood in syringe move into the tube by using pressure, so erythrocytes in the syringe are forced out through a small hollow needle causing ruptured membrane erythrocyte. Hemolysis characterized by the change of color of the serum to reddish. Objective: To determine the average of the result of the examination of AST enzyme levels in hemolyzed serum of 1 g/L and non- hemolyzed serum by using the IFCC kinetic method. Methods : The method of this research is Pre-Experimental with One Group Pretest-Postest research design . The method of examination used is IFCC kinetic. The research subjects were 36 people. The type of specimen used was hemolyzed serum of 1 g/L and non- hemolyzed serum. The result data of the examination were analyzed by using the Paired t-test statistical test . Results : The results showed that the average of AST level in hemolyzed serum of 1 g/L was 25.8400 U/L and non- hemolyzed serum was 24.9236 U/L. The results of statistical tests by using the Paired t-test showed a significant difference (Sig.2- tailed 0.039) between hemolyzed serum of 1 g/L and non- hemolyzed serum. Conclusion : Based on the results of the study, it can be concluded that there was a significant difference between AST enzyme levels in hemolyzed serum of 1 g/L and non- hemolyzed serum by using the IFCC kinetic method with a Sig.2-tailed value of 0.039 <0.05. Suggestion : The sample is used for AST examination to establish the diagnosis is non- hemolyzed serum. For further researchers, further research can be carried out regarding the examination of AST by using hemolyzed serum below of 1 g/L which is determined quantitatively and compared with non- hemolyzed serum, and before the examination, it is better to measure the serum pH first.
