Abstract

Platelet count is an examination that involves cases of hemostasis. Homogenization is an important stage in pre-analysis. After the blood is homogenized, there is usually a delay that causes the blood to settle, so it is necessary to do secondary homogenization so that the blood is homogeneous when it will be examined. In the field, there is no standard regarding secondary homogenization. Objective : To determine difference in the results of platelet count with secondary homogenization technique 5 and 8 times. Methods : This research use pre-experimental method with Static Group Comparison and Total Sampling sampling technique. Samples were homogenized primary 10 times and allowed to stand for 60 minutes, then samples were homogenized secondary 5 and 8 times. Platelet counts were checked using Sysmex KX-21. Data were analyzed by Paired Sample T-Test with 95% confidence level. Result : 5 times secondary homogenization has a mean value of 312.87⨉103/μL and 8 times secondary homogenization has a mean value of 315.03⨉103/μL. The data were distributed normally and continued with the hypothesis test, namely the Paired Sample T-Test and the probability results were 0.505>0.05, which is means that there was no difference. Conclusion : Based on the results of the study, it was concluded that there was no difference in the number of platelets in the secondary homogenization of 5 and 8 times inversion technique. Suggestion : For further research, it is recommended that the secondary homogenized platelet examination with the inversion technique 5 and 8 times can be left for more than 60 minutes.
