Abstract

Background :Hemoglobin examination is important to determine whether a person is anemic or not.Hemoglobin examination consists of 3 stages, namely pre-analytical, analytical and post-analytic.To get the right results, the three stages must be carried out according to the procedure.In the examination of hemoglobin, the blood used should not be frozen.To prevent clotting, K2EDTA anticoagulant was used and homogenization 10 times back and forth.EDTA blood when allowed to stand for a certain time blood cells will settle.deposition occurs in 3 stages, the first stage is the formation of rouleaux, the second stage is sedimentation and the stage is not compaction.Blood that has been allowed to stand will then be examined without secondary homogenization, the results of the examination are not correct. Objective :Knowing the results of hemoglobin examination in K2EDTA blood which was delayed 8 minutes and delayed 15 minutes after primary homogenization. Metode :This type of research is pre-experimental with Static Group Comparison and the sample population examined is 33.The subject's venous blood was taken as much as 4 cc, divided by 2 each by 2 per tube.Both tubes were primary homogenized 10 times.The first tube was allowed to stand for 8 minutes and the second tube was left for 15 minutes and then examined using Sysmex KX-21N.The data was analyzed using the Wilcoxon test with a 95% confidence level. Result :The hemoglobin level which was delayed 8 minutes after primary homogenization obtained an average result of 16.0 g/dL and a standard deviation (SD) of 1.6 g/dL.The hemoglobin level which was delayed 15 minutes after primary homogenization obtained an average result of 19,1 g/dL and a standard deviation (SD) of 2,0 g/dL.The statistical test results showed a significant difference of P = 0,000 (<0.05). Conclusion :Based on the results of the study concluded there are differences in the results of hemoglobin examination in blood that was delayed 8 minutes and 15 minutes after primary homogenization. Suggestion : Hemoglobin examination immediately after primary homogenization, and after a delay of 6 minutes without secondary homogenization Keywords : Hemoglobin, Delay, primary homogenization
