Abstract

Background: EDTA blood that has been allowed to stand for some time should be subjected to secondary homogenization, if it is to be examined. If the secondary is not homogenized the result will be false low. Therefore, an EDTA blood study was conducted without secondary homogenization and after secondary homogenization 8 times to prove there was a difference in leukocyte examination. Objective: To determine the results of the examination of the number of leukocytes in EDTA blood without secondary homogenization and after 8 times secondary homogenization. Methods: The type of research used is Pre-experimental with Statistical Group Comparation. 2 ml of venous blood was taken, the blood was allowed to stand for 10 minutes and then examined without secondary homogenization and after secondary homogenization 8 times with sysmex KX-21. The difference between the two results was analyzed using the Paired T test with a 95% confidence level. Results: From the 33 subjects examined, the average leukocyte count was 5.0 x10³/μL in EDTA blood without secondary homogenization and 7.2 x10³/μL in EDTA blood after 8 times secondary homogenization. The results of statistical tests showed that there was a significant difference = 0.000 (<0.05). Conclusion: There are differences in the number of leukocytes in EDTA blood without secondary homogenization and after 8 times secondary homogenization. Suggestion: Further research on the number of leukocytes in EDTA blood that is left for less than 10 minutes after primary homogenization.
