Abstract

Background: Platelet examination is important to determine a person's thrombocytosis or thrombocytopenia. Examination of the platelet count consists of 3 stages pre-analytical, analytical and post-analytical. To get the right results, the three stages must be carried out according to the procedure. On examination, the blood platelets used should not be frozen. To prevent clotting, the anticoagulant K2EDTA was used and homogenized 8 times back and forth. EDTA blood that is allowed to settle will experience precipitation which takes place within 10 minutes. Blood that has been allowed to stand if it is to be examined without secondary homogenization is not correct. Objective: To know the results of the examination of the EDTA blood platelet count without secondary homogenization and after 8 times secondary homogenization. Methods: This type of research is pre-experimental with Static Group Comparison and the sample population examined is 33. The subject's venous blood was examined using the Sysmex KX-21 instrument with the Impedance method. The research was conducted at the UKMC Fikes Laboratory. The sample used in this study was UKMC Fikes students who were taken by total sampling. The data were analyzed using the Paired T test. Results: On examination of the number of platelets without secondary homogenization the average result is 205.7 x 10³/µL, on examination of platelets after secondary homogenization 8 times the average result is 312.4 x 10³/µL Conclusion: Based on the research, it was concluded that the test results showed a significant difference in the number of EDTA blood platelets without secondary homogenization and after secondary homogenization 8 times with = 0.000 (<0.05). Suggestion: The study of examining the number of platelets in EDTA blood is left for less than 10 minutes. Keywords: Platelet Examination, homogenization
