Abstract

Background: Hematology laboratory examination consists of many kinds of examinations, one of the examinations carried out is leukocyte examination, the leukocyte examination is carried out primary homogenization by means of inversion 8 times and allowed to stand before examination. In this study using 8 and 15 minutes, this was done at an interval of 8 minutes when aggregation occurred in the formation of rouleaux. Samples that are allowed to stand for more than 10 minutes will experience precipitation. Objective: To determine the results of the examination of leukocytes in blood that was left for 8 and 15 minutes not to be secondary homogenized Methods: The type of research used is Pre-experimental with Statistical Group Comparation. Venous blood was taken 2 ml, primary homogenized 8 times after being left for 8 minutes not secondary homogenized and 15 minutes not secondary homogenized examined with sysmex KX 21. The difference between the two results was analyzed by Paired T test with 95% confidence level. Results: From the 33 results examined, the average value of the leukocyte count was 5.6 x10³/μL, the number of leukocytes that were left untreated for 8 minutes, and the average value of the number of leukocytes that were left untreated for 15 minutes was 4.3 x10³/μL. The test results showed the difference between the number of leukocytes in the blood that was left for 8 and 15 minutes not secondary to homogenization with = 0.000 (<0.05). Conclusion: Based on this study, it can be concluded that there are differences in the results of the examination of the number of leukocytes that were left for 8 and 15 minutes and were not secondary homogenized. Suggestion: In further research, add variables to be checked immediately, leave for 6 minutes and 12 minutes not to be secondary homogenized Keywords: Leukocyte Examination, Homogenization, Silent 8 and 15 minutes
