Abstract

There are three stages of laboratory examination, namely pre-analytical, analytical, and post-analytic. Sample processing is one of the pre-analytic stages. One of the factors that affect the results of the platelet count is homogenization of blood with anticoagulants. Currently, several laboratories have carried out homogenization using automated tools, but there are still some that are still manual. Objective : This study was to determine whether there is a difference in platelets count in K2EDTA blood samples which were homogenized secondary to manual inversion technique 10 times and automatic rolling technique for 1 and 2 minutes. Methods : This study was conducted on 31 subjects who met the inclusion and exclusion criteria with the type of pre-experimental research method "One Static Group Comparison". The research sample was given treatment, namely secondary homogenization manually with 10 times inversion technique, automatic rolling technique for 1 and 2 minutes, then platelet examination was carried out. Results : The results of the Wilcoxon test showed that there were differences in platelets count found in samples that were homogenized manually with 10 times inversion technique and 1 minute automatic rolling technique with a sig value of 0.008 (<0.05) and manual homogenization with 10 times inversion technique and automatic rolling 2 minutes technique. with a sig value of 0.000 (<0.05). The results of the Wilcoxon test showed that there was no difference in the number of platelets in the samples that were homogenized automatically using the 1 minute and 2 minute rolling technique with a sig value of 0.323 (> 0.05). Conclusion :There is a difference in platelets count in the sample homogenized by the manual inversion technique of 10 times with the automatic homogenization of the 1 minute and 2 minute rolling technique and there is no difference in the number of platelets in the sample which is automatically homogenized with the 1 minute and 2 minute rolling technique. Suggestion :Sample handling for platelet count examination can use the 10 times inversion technique and the next researcher can conduct a study with the same title, but the automatic homogenization time is accelerated to 30 seconds.
