Abstract
Laboratory examination goes through 3 stages, namely pre- analytical, analytical and post-analytic stages. One that is included in the pre- analytic stage is the sample homogenization process. Inadequate homogenization during analysis which causes clumping in the sample may result in false cell count results. Some sources suggest using manual homogenization inversion technique and other sources suggest automatic homogenization. This study aims to see whether there is a difference in the number of leukocytes that were homogenized secondary to manual 10 times inversion technique and automatic rolling technique for 1 minute and 2 minutes. Methods : This study includes a pre-experimental 3 variables with a statistical group comparison research design. Samples were taken using a total sampling technique that met the inclusion and exclusion criteria totaling 31 people. The leukocyte count was checked with homogenized samples manually using 10 times inversion technique and automatic rolling technique for 1 minute and 2 minutes. Research Results: The results of the leukocyte count that were homogenized with the 10 times inversion technique had an average of 7112 /ul, and the automatic rolling technique for 1 minute and 2 minutes had an average of 7003 /ul and 7093 / ul. After the Repeated Anova test, p-values were obtained respectively 0.791, 1,000, and 0.620 p>0.05, which means there is no difference. Conclusion : Based on this study, it can be concluded that there is no difference in the number of leukocytes that were homogenized secondary to manual inversion technique hp times and automatic for 1 minute and 2 minutes. Suggestion : Future researchers can conduct research using other homogenization tools such as rotary tipe.
