Abstract

Background : Antibiotic susceptibility testing is examination to determine the bacteria that disease. One factor that must be considered is the incubation temperature. CLSI sets the standard incubation temperature at 37°C. Escherichia coli is a mesophilic bacterium that thrives at temperatures 30°C and 37°C. The Meteorology, Climatology, and Geophysics Agency stated that the temperature in Palembang in May 2026 will be 23°C and 31°C. Therefore, incubation at room temperature can be an alternative for antibiotic susceptibility testing of Escherichia coli. Objective : This compared the inhibition zones of Escherichia coli bacteria using incubation at non-air-conditioned room temperature, air-conditioned room temperature, and incubator temperature. Method: This type of pre-experimental study with a Statistical Group Comparison design. The study was conducted in the Microbiology Laboratory of the Faculty of Health Sciences, Musi Charitas Catholic University. The study included 30 data sets. Data analysis used the Tukey Post Hoc Test. Results: The average inhibition zone diameter of Escherichia coli in incubation at non-ac room temperature was 36.03 mm, at AC room temperature 35.99 mm, and at incubator temperature 36.08 mm. The results of the One-Way ANOVA test showed a significance of 0.961 (p>0.05), indicating no significant difference between the treatments. Conclusion: The use of incubation at non-ac room temperature and ac room temperature for Tetracycline antibiotic susceptibility testing of Escherichia coli can be used as an alternative to incubator incubation. Suggestion: Room temperature incubation can be used as alternative incubation, future researchers need to maintain the temperature of the air-conditioned room to keep it stable.
