Abstract

Background: Blood grouping requires high-quality antisera to produce accurate agglutination reactions. Antisera contain specific antibodies that react with antigens on the surface of erythrocytes. Reagent quality may be affected by storage temperature and duration. Objective: To determine differences in agglutination grades in blood grouping using the tube method with antisera stored for 0, 5, 6, and 7 days at 2–8°C. Methods: This study used an analytical observational design with a cross-sectional approach and repeated measurements on the same samples. A total of 30 subjects were included. Testing was performed using 5% erythrocyte suspensions reacted with anti-A, anti-B, and anti-AB antisera stored for 0, 5, 6, and 7 days. Conclusion: Antisera stored at 2–8°C maintained optimal agglutination reactions for up to seven days in blood grouping using the tube method,agglutination grade was 4+ for all storage durations. The Kruskal–Wallis test yielded a p-value of 1.000 (p > 0.05), indicating no significant difference in agglutination grades among antisera stored for 0, 5, 6, and 7 days. . Recommendation: Antisera stored for up to seven days at 2–8°C may be used for blood grouping by the tube method, provided that storage conditions and reagent quality control are properly maintained.
