Abstract
Background: Escherichia coli is a common cause of infection, and its susceptibility to antibiotics is frequently tested using the Kirby-Bauer disk diffusion method. Tetracycline is a broad-spectrum antibiotic that effectively inhibits the growth of E. coli. Susceptibility testing can be influenced by technical factors, specifically the handling of antibiotic disks, which must be acclimated from storage temperature to room temperature to prevent condensation. The literature specifies a standard acclimatization time of 60 minutes. This timeframe is inefficient in laboratories with a high sample volume, necessitating a more efficient acclimatization time without compromising test results. Objective: To determine the effect of varying acclimatization times for tetracycline antibiotic discs on the inhibition zone diameter of E. coli. Methods: A pre-experimental study using a Static Group Comparison design. The samples consisted of pure E. coli ATCC 25922 strains treated with tetracycline. Based on Federer’s formula, each treatment was repeated 10 times. Data were analyzed using the Kruskal-Wallis test followed by the Dunn post hoc test. Results: The average diameter of the inhibition zone was 26.8 mm after 5 minutes of acclimatization, 26.8 mm after 10 minutes, 27.2 mm after 15 minutes, 27.1 mm after 30 minutes, and 28.3 mm after 60 minutes. The Kruskal-Wallis test showed p=0.012. The Dunn post hoc test for 5 minutes was p=0.020; for 10 minutes, p=0.019 (p<0.05); for 15 minutes, p=0.481; and for 30 minutes, p=0.267 (p>0.05). Conclusion: There is a significant effect of varying acclimation times (5 and 10 minutes) for the tetracycline antibiotic disk on the inhibition zone diameter of E. coli (p = 0.012). The most efficient and optimal acclimation time is 15 minutes. Suggestion: The acclimation time for antibiotic discs should be 15 minutes. Further research is recommended using different types of antibiotics and bacterial species.
