Abstract

Background: Blood typing is performed to determine blood group type, with the tube method serving as the gold standard, which includes serum grouping and cell grouping using a 5% erythrocyte suspension. Test results can be influenced by erythrocyte concentration and the red blood cell washing process using a 0.9% NaCl solution. Objective: To determine differences in the degree of agglutination in tube-based blood typing using red blood cell suspensions that were rewashed and those that were not rewashed after 24 hours of storage at 2–8°C. Methods: An analytical observational study with a cross-sectional design. The sample consisted of 5% red blood cell suspensions from 33 students in the DIV Medical Laboratory Technology program. Testing was performed after 24 hours of storage. Data were analyzed using the Wilcoxon signed-rank test. Results: Of the 33 samples with two different treatments, the results using a 5% red blood cell suspension-both with and without rewashing after 24 hours of storage-showed a 4+ agglutination level in all samples. Conclusion: Based on the study results, it was concluded that there was no difference in the degree of agglutination during blood typing using a 5% RBC suspension without rewashing and with rewashing. The results of the Wilcoxon Signed-Rank Test showed a p-value of 1.000 (>0.05). Recommendation: A 5% RBC suspension without rewashing remains suitable for tube blood typing after 24 hours of storage at 2–8°C.
