Abstract

Background: There are several types of clinical laboratory tests, one of which is blood type testing. In the ABO blood type examination using the tube method with red blood cell suspension, the commonly used erythrocyte suspension is 2-5% erythrocyte suspension. In the manufacture of 5% cell suspension that the time required in making the suspension ± 30 minutes to 1 hour from the washing stage to the suspension of blood cells. So that the manufacture of blood cell suspension by washing has the same degree of agglutination result as the manufacture of cell suspension without washing. Purpose: This study aims to determine the degree of agglutination in blood type examination using 5% red blood cell suspension with washing and 10% blood cell suspension without washing. Method: The type of research that will be used is observational analytical research with a cross sectional design. The research subjects used were 38 DIV students of medical laboratory technology. Furthermore, statistical analysis was carried out using the wilcoxon signed rank differential test. Result: From 38 samples with two different treatments, the results of using a red blood cell suspension of 5% with washing and 10% without washing were obtained from 13 samples of blood type A, 14 samples of blood type B, and 11 samples of blood type O. The results of the average degree of agglutination showed 4 positives (4+) for blood types A and B. Conclusion: Based on the results of the study, it was concluded that there was no difference in the results of blood type examination using 5% HR suspension with washing and 10% HR without washing. A value (sig 2 tailed) was obtained >0.05. Suggestion: A 10% red blood cell suspension without washing can be considered for use as a rapid examination material.
