Abstract

Background : Clinical Chemistry examination consists of many types of examination, one of the examinations carried out is the Gamma Glutamyl Transferase (GGT) enzyme examination. Laboratory Internal Quality Assurance has three phases, namely pre-analytical, analytical and post-analytical stages. At the analytical stage, one of the most important laboratory examinations is the selection of reagents. Due to the renewal of reagents with different reagent compositions, wavelengths and calibrators, there is a comparison of the results of the GGT enzyme examination itself. So in this study we wanted to compare the results of the GGT enzyme using GGt1 and GGT2 (Six Sigma Strong) reagents on the Architect Tool. Objective : To find out whether the GGT enzyme results match between GGT1 and GGT2 Reagents (Six Sigma Strong) in serum samples. Method : The type of research used was analytical observational with a cross sectional design. Serum was examined for the GGT enzyme with GGT1 and GGT2 Reagents (Six Sigma Strong) on an Architect C8000 instrument. The differences between the two results were analyzed using the Comparative Numerical Conformity Test (Bland Altman) with a confidence level of 95% and the Limit of Agreement was between -5 and +5. Results : Of the 50 serum samples examined, the average value of the GGT enzyme using the GGT1 reagent was 85.04 U/L with the lowest result being 11 U/L and the highest result being 202 U/L, while the average value using the GGt2 reagent was 85.00 U/L. L with the lowest yield of 13 U/L and the highest yield of 200 U/L. These results show that 98% of the GGT enzyme results are within the acceptance limits with a Mean Difference of 0.040, lower limit of agreement -4.1499 and upper limit of agreement 4.2299. Conclusion : There is suitability/reliability in the results of measuring the GGT enzyme using GGT1 and GGT2 reagents on the Architect C8000 instrument. This research shows that both reagents can be used interchangeably on the Architect C8000 device. Suggestion : In the next research, a comparative test of reagents with different parameters was carried out using the Architect tool.
